Potential Role of Manila Sapodilla (Manilkara zapota van Royen Syn. Achras zapota L.) Leaf Subfractions Prepared by Radial Chromatography Against Pseudomonas aeruginosa Bacteria

Pseudomonas aeruginosa is a pathogenic bacterium frequently implicated in skin infections, particularly in abrasions, wounds, and burns. This study aimed to evaluate the antibacterial potential of the extract and subfraction of manila sapodilla leaves against Pseudomonas aeruginosa, and to compare their activity at concentrations of 5%, 10%, and 15%. Extraction was carried out by maceration using 96% ethanol, followed by liquid–liquid fractionation with n-hexane, ethyl acetate, and water. The fraction was further purified using radial chromatography to obtain subfractions. Phytochemical screening and thin-layer chromatography confirmed the presence of flavonoids in both the extract and subfraction. Antibacterial activity was tested using the disk diffusion method, with Pseudomonas aeruginosa as the test organism, 10% DMSO as a negative control, and 0.01% Ciprofloxacin as a positive control. The results revealed that both extract and subfraction exhibited antibacterial activity, with inhibition zone diameters increasing in proportion concentration. Statistical analysis using two-way ANOVA indicated significant differences (p < 0.05) between concentrations and sample types. Therefore, these findings indicate that Manilkara zapota leaf extract and subfractions possess potential as natural antibacterial agents against Pseudomonas aeruginosa.
